چکیده

Introduction It is now appropriate to discuss fine details of the mechanism of action of xanthine oxidase because of the extensive spectroscopic and kinetic evidence available; this is especially so since this has recently been supplemented by X-raycrystallographic data. Much of the information has been reviewed comprehensively [1,2] and is also discussed in this volume [3,4]. In this article we intend to concentrate on the role of electron nuclear double resonance (ENDOR) spectroscopy in determining aspects of the structure of intermediates in the mechanism, with particular reference to details of the evidence for Mo-C bonds, since there may be misunderstandings in the literature concerning how we interpreted our results [S]. We go on to consider some of the various hypotheses concerning the mechanisms that are currently being debated.

Molybdenum EXAFS of the Desulfovibrio gigas Mo(2Fe-2S) protein--structural similarity to "desulfo" xanthine dehydrogenase.

The molybdenum EXAFS of the Mo(2Fe-2S) protein from Desulfovibrio gigas has been examined using fluorescence detection and synchrotron radiation. In the oxidized form the molybdenum environment is found to contain two terminal oxo groups and two long (2.47 A) Mo-S bonds. Evidence was also found for an oxygen or nitrogen donor ligand at 1.90 A. Addition of dithionite to the oxidized enzyme resul...

Summer Savory (Satureja hortensis) Extract Inhibits Xanthine Oxidase

Xanthine oxidase plays crucial roles in the production of reactive oxygen species (ROS), and uric acid in blood which may lead to gout, one of the oldest known forms of arthritis amongst humans. Summer savory (Satureja hortensis L.) a medicinal/spice exhibits antioxidant activity, but unknown effects on xanthine oxidase activity. Here, for the first time, we examined the effects of S. hortensis...

Inhibitory effect of Physalis alkekengi extracts in different phenological stages on xanthine oxidase activity

Introduction: Physalis alkekengi (Solanaceae) is a rich source of various antioxidants. There are some reports that show P. alkekengi has been used for treatment of a wide range of diseases including gout and inflammation. Xanthine oxidase plays a crucial role in gout. Many natural compounds such as various flavonoids have been reported to have inhibitory effect on xanthine oxidase. Metho...

Arsenite-Inhibited Xanthine Oxidase-Determination of the Mo-S-As Geometry by EXAFS

The interaction of arsenite with oxidized, partially reduced, and fully reduced forms of xanthine oxidase has been studied by X-ray absorption spectroscopy at the arsenic and molybdenum K edges. Clear evidence for a Mo-As interaction at 3.00 8, is observed in the molybdenum EXAFS of the fully reduced ternary complex consisting of xanthine oxidase, arsenite, and the inhibitor 8-bromoxanthine. An...

Onion, a Potent Inhibitor of Xantine Oxidase

Onion (Allium Cepa) contains high levels of flavonoids. Although there are many studies indicating the inhibitory effects of flavonoids on xanthine oxidase, there is no report on the effect of onion on this enzyme. Therefore, in the present study, the inhibitory effects of onion on xanthine oxidase are investigated. Fresh filtered juice of onion was prepared and its inhibitory effect on guine...
